Development of Infectious Clones of Sugarcane Mosaic Virus

Abstract: 

Sugarcane mosaic virus (SCMV), a member of the genus Potyvirus, family Potyviridae, infects maize and is non-persistently transmitted by aphids. We report development of full-length infectious clones of two isolates of SCMV, one from Ohio (SCMV-OH) and one from Germany (SCMV-GR). SCMV-GR has 79% in nucleotide and 89% in amino acid sequence identity to SCMV-OH with most sequence variability in P1 and N-terminus of coat protein regions. Previous report demonstrated SCMV-GR was not able to infect some maize breeding lines susceptible to SCMV-OH. Development of the infectious clones allows us to next determine the molecular basis of differential host range or resistance-breaking of SCMV-OH.
